In ELISA testing, positive and negative controls are essential for ensuring the accuracy and reliability of results. These control samples not only validate the test procedure but also serve as reference points to interpret the outcomes. To maintain consistency, the composition of these controls—especially the negative control—should closely resemble that of the actual test sample. When working with human serum as the specimen, it is ideal to use human serum as the control material, as normal serum can generate varying background signals depending on the ELISA format used. However, due to the difficulty in obtaining large quantities of normal human serum, many foreign ELISA kits use recalcified human plasma as a base. This involves adding calcium ions to plasma, allowing fibrinogen to form a clot, which is then removed. The resulting liquid has a composition similar to serum and serves as an effective control.
The negative control should contain no target analyte, such as HBsAg, and ideally should also be negative for other related markers like anti-HBs. On the other hand, the positive control typically contains a buffer solution with a protein stabilizer and a known quantity of the target substance. The concentration of this substance is usually specified in the kit’s instructions and should match the sensitivity of the reagent. During testing, the absorbance values from the positive control are compared with those of the test samples, allowing for an approximate estimation of the analyte levels in the specimens. For example, if the ELISA kit detects HBsAg with a sensitivity of about 0.5 ng/ml, the positive control may contain around 10 ng/ml of HBsAg.
To ensure long-term stability, preservatives and antibiotics are often included in the control solutions. Additionally, quantitative ELISA kits must include a standard curve, which requires at least four to five different concentrations spanning the detectable range. These standards are usually prepared in a buffer containing protein protectants and preservatives to maintain their integrity over time. Properly designed controls and standards are crucial for accurate and reproducible ELISA results, making them indispensable components of any reliable immunoassay.

Angle brushes are filaments that have been cut on an angle – these are excellent to use in small or curved areas of the painting.Filberts Brushes with oval shaped filaments are known as filberts in smaller sizes and oval wash in larger sizes.
