The detection method of Sudan red dye in Chili powder is as follows:
1. Scope of application: suitable for the detection of Sudan red dye in chili oil and chili powder foods
2. Sample pretreatment
2.1 Weigh 1 g to 5 g (accurate to 0.01 g) of powdered samples such as red pepper in a triangular flask, add 10 mL to 30 mL of n-hexane, sonicate for 5 min, filter, and wash the residue several times with 10 mL of n-hexane to the eluent Colorless, combined with n-hexane solution, concentrated to less than 5mL with a rotary evaporator, and slowly added to Welchrom ® Alumina-N (500mg / 3mL, part No. WSAN010305) column. When the liquid level is about 2mm, the sample should be loaded. The column should not be dried during the entire chromatography process. The concentrated bottle is rinsed with n-hexane a few times and injected into the chromatography column. The band width of the pigment to be adsorbed on the surface of the alumina should be less than 0.5cm. After the sample solution completely flows out, depending on the amount of oily impurities in the sample, wash the column with 10mL-30mL n-hexane until the effluent is colorless. The liquid was eluted with 60 mL of n-hexane solution containing 10% acetone. After collecting and concentrating, transfer to a volume of 5 mL with acetone and dilute to 0.4 mL with a 0.45 μm organic filter membrane.
2.2 Oily samples such as red pepper oil, hot pot ingredients, cream, etc. Weigh 0.5 g ~ 2g (accurate to 0.01g) sample in a small beaker, add appropriate amount of n-hexane to dissolve (about 1 mL ~ 10mL), difficult to dissolve samples Warm and dissolve in the alkane. Press "Add slowly to Welchrom ® Alumina-N column ... to be tested after filtration" in 2.1.
3. Chromatographic conditions Column: Ultimate XB-C18 250 * 4.6mm, 5µm
Mobile phase: acetonitrile / water solution, gradient elution;
Flow rate: 1.0ml / min
Column temperature: 40 ℃
Injection volume: 20µL
Detector: UV 223nm
